About | The Somru® PEG ELISA kit is a competitive assay has been validated for the quantitation of wide variety of free and conjugated PEG (linear or branched) molecules (>5 kd sizes) in serum or plasma. The assay is specific to the backbone of polyethylene glycol (PEG). |
Detection method | Colorimetric; absorbance at 450 nm with reference absorbance at 650 nm. |
Storage | Store the unopened kit at 2-8 °C. |
Safety Warnings and Precautions | All chemicals should be considered potentially hazardous. We recommend that this product and its components be handled by those trained in laboratory techniques and be used according to the principles of Good Laboratory Practice. |
Quality Control Samples (QCs) | Three levels of QCs are included for custom GLP kits for an additional cost. QCs are prepared by fortifying 20 kD linear PEG in human plasma or serum. |
Precision | The precision was determined by analyzing samples prepared at 40 ng/mL of 20 kD linear PEG in 6 replicates on 6 different occasions. Intra-assay precision (coefficient of variation(CV)) ranges from 8% to 14%. Inter-assay precision was at 14%. |
Accuracy/ Recovery | 40 ng/mL of PEG (20 kD linear) was fortified in 10 lots of human serum. Recovery ranges are from 94-119% with an average recovery of 111%. |
Total Error | Total Error (TE) was calculated by adding the mean %bias with inter-assay variability (%CV). The TE was found to be less than 21%. |
Sensitivity | The limit of detection is 0.7 ng/mL. |
Dilutional Linearity | Tested up to 100 µg/mL |
Hook Effect | No hook effect was observed up to 100 µg/mL of PEG. |
Interferences | Polyetheylene derivatives such as Tween, Triton, NP‐40 etc. interferes with this assay due to their structural similarity to PEG and should not be used in this assay. |
Sample Pre-treatment (ADA Interference) | If a high level of anti-PEG antibody is expected to be present in test samples, an acid dissociation step may be incorporated to minimize the impact of these antibodies in PEG quantitation.
