| About | The Somru® mPEG ELISA kit is an enzyme immunoassay for the quantitative determination of mPEG and mPEGylated proteins in serum or plasma. This user-friendly assay design utilizes a pair of antibodies that allows the detection of various PEG sizes with methoxy terminal. This kit was calibrated using 20kD mPEG. |
| Detection method | Colorimetric; absorbance at 450 nm with reference absorbance at 650 nm. |
| Storage | Store the unopened kit at 2-8 °C. |
| Quality Control Samples (QCs) | Three levels of QCs are included with custom GLP kits for an additional cost. QCs are prepared by fortifying 20kD mPEG in human serum or plasma. |
| Precision
|
The precision was determined by analyzing samples prepared at 10 ng/mL in 6 replicates on 6 different occasions. Intra-assay precision (coefficient of variation(CV)) ranges from 11% to 15%. Inter-assay precision was at 16%. |
| Accuracy | The accuracy was determined by analyzing three levels of quality control samples in 6 replicates on 6 different occasions. Mean %bias was less than 19%. |
| Total Error | Total Error (TE) was calculated by adding the mean %bias with inter-assay variability (%CV). The TE was found to be less than 25%. |
| Sensitivity | The detection limit is 0.19 ng/mL |
| Dilutional Linearity | Dilutional linearity was established up to 1,000 fold. Samples with concentration up to 1000 µg/mL can be measured in this assay. |
| Sample Pre-treatment (ADA Interference) | If a high level of anti-PEG antibody is expected to be present in test samples, an acid dissociation step may be incorporated to minimize the impact of these antibodies in etanercept quantitation. | Comments | Do not substitute user-prepared buffers for those provided with the kit (i.e. diluent or wash buffer). Many commercially available materials and buffer contain PEG or PEGylated molecules and this will affect performance of the kit. For this reason, also, care should be taken when selecting other components used in this assay. |
